بررسی مقاومت ژنتیکی گوسفندان نژاد قزل و آمیخته قزل-بلوچی نسبت به نماتودهای دستگاه گوارش
زمینه مطالعاتی: امروزه در کنار صفات تولیدی، ویژگیهای مقاومت ژنتیکی دام نیز مورد توجه اصلاح گران قرار گرفته است و علاقمندی به استفاده از قابلیتهای ژنتیکی حیوان برای مقابله با بیماریها و انگلها فزونی یافته است. هدف: هدف از اجرای این طرح، بررسی مقاومت نسبی گوسفندان نژاد قزل و دو رگه های قزل- بلوچی در استان آذربایجان شرقی نسبت به انگلهای داخلی بود. روش کار: اطلاعات تحقیق از 24 رأس گوسفند یکساله حاصل از دو ترکیب ژنتیکی بدست آمد. گروههای ژنتیکی شامل 12 رأس گوسفند نژاد قزل و 12 رأس گوسفند آمیختهی قزل-بلوچی بود. نمونه گیری در دو نوبت و در تاریخهای 30/11/1390 و 14/12/1390 در ایستگاه تحقیقاتی خلعت پوشان انجام گرفت. تمامی گوسفندان متولد زمستان 1389 بودند و همهی آنها در شرایط محیطی یکسان قرار داشتند. صفات مورد بررسی شامل وزن بدن، تعداد تخم نماتودها در هر گرم مدفوع، هماتوکریت و تست فماچا بودند. داده های وزن تولد و هماتوکریت توسط رویهی GLM و بقیه صفات توسط رویهی MIXED نرم افزارSAS آنالیز شد. نتایج: نتایج بدست آمده نشان داد که اثر گروه ژنتیکی در مورد کل تخم نماتودهای موجود در هر گرم مدفوع، تست فماچا و هماتوکریت ، تخم نماتودیروس و مارشالاژیا معنیدار است(P<0.05). دورگ گیری قزل با بلوچی موجب افزایش مقاومت نسبی به نماتودهای دستگاه گوارش از لحاظ صفات اندازه گیری شده در این تحقیق است ولی دلیل ژنتیکی برتری دورگ ها نسبت به نژاد خالص و آگاهی از ماهیت پلی ژنی یا اثرات غیر افزایشی نیاز به تحقیقات بیشتر دارد.

تاثیر پودر سیر و آنتی بیوتیک فلاوومایسین بر عملکرد جوجههای گوشتی تغذیه شده با جیرههای حاوی سطوح متفاوت چربی خام
زمینه مطالعاتی: سطح چربی جیره ممکن است روی تاثیرگذاری گیاهان داروئی و عصاره آنها بر عملکرد طیور موثر باشد. هدف: این تحقیق به منظور ارزیابی تاثیر پودر سیر و آنتیبیوتیک فلاوومایسین بر عملکرد و قابلیت هضم چربی جوجههای گوشتی تغذیه شده با سطوح متفاوت چربی انجام گرفت. روش کار: سیصد قطعه جوجه گوشتی سویه راس 308 با آرایش فاکتوریل (2×3) با 2 سطح چربی خام جیره و 3 نوع افزودنی در قالب طرح بلوکهای کامل تصادفی به 6 تیمار، 5 تکرار و 10 پرنده در هر تکرار اختصاص داده شدند. جیرههای آزمایشی عبارت بودند از 1) جیره پایه (حاوی 3% چربی)، 2) جیره پایه بعلاوه 2% چربی (جیره 5% چربی)، 3) جیره پایه بعلاوه 5/1% پودر سیر، 4) جیره 5% چربی بعلاوه 5/1% پودر سیر، 5) جیره پایه بعلاوه فلاوومایسین (200 گرم در تن) و 6) جیره 5% چربی بعلاوه فلاوومایسین (200 گرم در تن). نتایج: نتایج آزمایش نشان داد که جیرههای آزمایشی تاثیر معنیداری بر خوراک مصرفی، ضریب تبدیل خوراک، قابلیت هضم چربی، متابولیتهای خونی، درصد اجزای لاشه و جمعیت سلولهای خونی جوجههای گوشتی نداشتند (05/0P > ) اما جوجههای دریافت کننده جیره پایه همراه با آنتیبیوتیک فلاوومایسین یا جیره حاوی مکمل چربی همراه با پودر سیر دارای افزایش وزن بدن بالاتری نسبت به گروه شاهد بودند (05/0P < ). جوجههای دریافت کننده جیره دارای مکمل چربی دارای IgG اولیه و کل بالاتری نبست به جوجههای دریافت کننده جیره پایه بودند (05/0P < ). همچنین جوجههای دریافت کننده جیرههای حاوی پودر سیر دارای IgM و آنتیبادی کل اولیه و ثانویه و همچنین IgG ثانویه بالاتری نسبت به گره شاهد بودند (05/0P < ). نتیجهگیری نهایی: در مجموع، سطح چربی جیره موجب کاهش معنیدار خوراک مصرفی جوجههای گوشتی شد ولیکن پودر سیر موجب بهبود پاسخ ایمنی هومورال جوجههای گوشتی گردید.

جداسازی و کشت سلولهای اسپرماتوگونی گاو و بررسی اثر فاکتور رشد اپیدرمال بر تعداد سلولها، اندازه کلونیها و میزان زندهمانی اسپرماتوگونی
زمینه مطالعاتی: فاکتور رشد اپیدرمال میتواند باعث افزایش تعداد سلولها، اندازه کلونیها و میزان زندهمانی سلولهای بنیادی اسپرماتوگونی گردد. هدف: این آزمایش به منظور بررسی اثر فاکتور رشد اپیدرمال بر تعداد سلولها، اندازه کلونیها و میزان زندهمانی اسپرماتوگونی انجام گرفت. روشکار: در این مطالعه سلولهای اپیتلیوم لولههای منیساز از بیضه گوساله با استفاده از مراحل هضم آنزیمی و DSA لکتین جداسازی شدند. ماهیت سلولها علاوه بر ریختشناسی و فعالیت آنزیم آلکالین فسفاتاز، از طریق نشانگرهای اختصاصی Oct-4 و ویمنتین در سلولهای کلونی و سرتولی تأیید شدند. برای تعیین شرایط مناسب کشت و غنیسازی سلولها، تعلیق سلولی حاوی سلولهای بنیادی به صورت هم کشت با سلول سرتولی و افزودن غلظتهای متفاوت از فاکتور رشد EGF کشت داده شد. در طول 2 هفته دوره کشت، میزان کلونیزاسیون، با میکروسکوپ نوری اندازه گیری شد. در آخرین مرحله، این سلولها در محیط In vitro منجمد-ذوب شدند تا درجه خلوص و قدرت زیست سلولهای بنیادی ارزیابی شود. نتایج: در بررسی سطح به صورت کلی، در تیمار با غلظت 50 نانوگرم EGF افزایش سطح تغییر معنیدار بود (05/0P <)، در بررسی کلونیهای با قطر بزرگتر از 114/0 میلیمتر (± SE)، افزایش قطر کلونیها در غلظت 50 نانوگرم معنیدار بود (05/0P <)، اما در اندازه کلونیها در شمارش روز آخر تغییر معنیداری در هیچ کدام از گروه های آزمایشی دیده نشد (05/0 P ≥). نتیجهگیری نهایی: مطالعه حاضر نشان میدهد که میتوان سلولهای بنیادی اسپرماتوگونی را با درجه خلوص بالا از بیضه گوساله جدا کرد و همچنین از رابطه متقابل بین سلولهای بنیادی اسپرماتوگونی و سلولهای سرتولی و بعضی فاکتورهای رشد برای شروع، حفظ فرایند اسپرمزایی و غنیسازی سلولهای بنیادی در طی کشت و افزایش درجه خلوص و قدرت زیست آنها در طی انجماد استفاده کرد. درتجزیه و تحلیل آماری از آزمون ANOVA استفاده شد و P <0.05، به عنوان سطح معنیدار در نظر گرفته شد.

اثرهای استفاده از اسانس ترکیبی گیاهان دارویی در آب آشامیدنی بر عملکرد رشد، هماتولوژی و پروفیل چربی خون شترمرغ
زمینه مطالعاتی: اسانسها به عنوان محرکهای رشد نقش مهمی در بهبود سلامتی در گونههای مختلف طیور دارند، اما در مورد تأثیر این فراوردههای گیاهان دارویی مطالعه خاصی روی جوجه شترمرغها انجام نشده است. هدف: در این مطالعه اثرهای استفاده از سطوح مختلف مکمل اسانس ترکیبی گیاهان دارویی (شامل سطوح مساوی از اسانس نعناع، اکالیپتوس، رازیانه و آویشن) بر عملکرد رشد، هماتولوژی و پروفایل چربی خون در جوجه شترمرغ اهلی آفریقایی از سن 5 تا 7 ماه مورد بررسی قرار گرفت. روش کار: سه تیمار آزمایشی عبارت بودند از: شاهد (بدون اسانس)، و سطح 200 و 400 میلیگرم اسانس در لیتر آب آشامیدنی. در مجموع 18 قطعه جوجه شترمرغ (6 پرنده برای هر تیمار) در این آزمایش مورد استفاده قرار گرفت. نتایج: سطح 200 میلیگرم از اسانس به طور معنیداری وزن بدن و ضریب تبدیل غذایی را از 5 تا 7 ماهگی بهبود بخشید (05/0P<). سطح 200 میلیگرم از اسانس همچنین سبب افزایش معنیدار غلظت هموگلوبین خون شد (038/0P=). علاوه بر این، افزودن هر دو سطح 200 و 400 میلیگرم اسانس در هر لیتر آب موجب کاهش معنیدار (05/0P<) در نسبت هتروفیل به لنفوسیت، به عنوان یک شاخص تنش گردید. هیچ اثر معنیداری از جیرههای آزمایشی بر روی دیگر فراسنجههای هماتولوژیکی مشاهده نشد (05/0<P). سطح 200 میلیگرم اسانس سبب کاهش معنیدار در سطح کلسترول سرم خون گردید (052/0P=). با این حال، مقادیر تری گلیسیرید، لیپوپروتئین با چگالی بسیار کم، لیپوپروتئین با چگالی بالا و لیپوپروتئین با چگالی کم خون تحت تاثیر تیمارهای آزمایشی قرار نگرفت (05/0<P). نتیجهگیری نهایی: به طور کلی، استفاده از غلظت 200 میلیگرم اسانس ترکیبی در جوجههای شترمرغ برای بهبود رشد، مقاومت به تنش و متابولیسم کلسترول موثر بود.

اثرات استفاده از دو هورمون eCG و GnRH بر عملکرد تولیدمثلی بزهای خلخالی در فصل تولید مثلی
زمینهمطالعاتی: افرایش نرخ تخمکگذاری در بزهای ماده منجربه بهبود عملکرد تولیدمثلی از طرق افزایش در نرخ دوقلوزایی و نرخ بزغالهزایی میگردد. هدف: هدف تحقیق حاضر بهبود عملکرد تولیدمثلی بزهای ماده با استفاده از هورمونهای GnRH و eCG بود.روش کار: برای این منظور تعداد 150 راس بز ماده نژاد خلخالی در قالب طرح کاملا تصادفی به سه گروه تقسیم شدند. برنامه همزمانسازی فحلی شامل اسفنجگذاری بمدت 16 روز بود. بعد از خارج نمودن اسفنجها، گروه کنترل (T1) بدون دریافت هیچ هورمونی به منظور جفتگیری در معرض بزهای نر قرار داده شدند. گروه دوم (T2) و گروه سوم (T3) به ترتیب 400 واحد بین المللی eCG و 400 واحد بین المللی eCG همرا با 50 میکروگرم GnRH به صورت عضلانی دریافت کردند. سپس بزها فحل یابی شدند و با بزهای نر جفتگیری کردند. همچنین غلظت هورمونهای استروژن و پروژسترون در زمانهای مختلف سیدر گذاری در گروهها اندازهگیری شد. نتایج: تیمارها تأثیری روی نرخ زایش و نرخ آبستنی بزها در فصل تولید مثلی نداشت (05/0< P). از نظر آماری نرخ بزغالهزایی در گروه کنترل (106 درصد) نسبت به گروههای دیگر (تیمارهای دوم و سوم به ترتیب 158 و 188) کمتر بود. همچنین راندمان تولید بزغاله در تیمارهای 1 تا 3 به ترتیب 113، 175 و 188 درصد محاسبه شد که از این نظر نیز، تیمارهای گروه آزمایشی نسبت به گروه کنترل اختلاف معنیداری در سطح 05/0 داشتند. نرخ دوقلوزایی در تیمارهای دوم و سوم به ترتیب 49 و 56 درصد بود که نسبت به گروه شاهد (13 درصد) تفاوت معنیداری در سطح 05/0 داشتند. غلظت هورمونهای استروژن و پروژسترون بین سه گروه آزمایشی در زمانهای مختلف سیدر گذاری اختلاف معنیداری نداشتند (05/0< P). تیجهگیری نهایی: استفاده از تیمارهای هورمونی در فصل تولید مثل در بز خلخالی عملکرد تولیدمثل را بهبود داد.

تعیین دما و pH بهینه رشد باکتریهای تجزیه کننده مواد لیگنوسلولزی جدا شده از دستگاه گوارش موریانه و اثر آنها بر قابلیت هضم و فراسنجههای تخمیر آزمایشگاهی برخی پسماندهای کشاورزی
زمینه مطالعاتی: روده موریانهها حاوی انواع مختلفی از میکروارگانیسمهای تجزیه کننده مواد لیگنوسلولزی است که امکان استفاده از آنها در بهبود ارزش تغذیهای پسماندهای زراعی گامی بزرگ جهت پوشش دادن کمبود خوراک دام است. هدف: این پژوهش به منظور تعیین دما و pH بهینه رشد باکتریهای همزیست تجزیه کننده مواد لیگنوسلولزی جدا شده از دستگاه گوارش موریانهها و بررسی قابلیت هضم و فراسنجههای تخمیر آزمایشگاهی کاه گندم و سرشاخه خرمای فرآوری شده با ایزولههای مذکور انجام شد. روشکار: برای این منظور، از سه گونه باکتری Bacillus licheniformis ، Ochrobactrum intermedium و Microbacterium paludicola، استفاده گردید که باکتریهای مذکور توسط کشت محتویات روده موریانهها در محیطهای کشت حاوی انواع مختلفی از لیگنین و مواد لیگنوسلولزی جداسازی شده بودند. منحنیهای رشد ایزولهها در محیط کشت نوترینت براث به دست آمد. و سپس کاه گندم و سرشاخه خرما توسط جدایههای مذکور عملآوری شدند. نتایج: رشد بهینه باکتری B. licheniformisدر دمای 40 درجه سانتیگراد بود، اما در دو گونه O. intermedium و M. paludicola رشد بهینه در دمای 37 درجه سانتیگراد مشاهده گردید. رشد بهینه هر سه سویه در pH برابر با 7 اتفاق افتاد. پس از عملآوری، برای هر دو سوبسترای کاه گندم و سرشاخه خرما بیشترین حجم گاز تولیدی و قابلیت هضم ماده آلی در تیمار فرآوری شده توسط باکتری B. licheniformisو کمترین در تیمار شاهد مشاهده گردید (05/0P<). قابلیت هضم ماده خشک، سوبسترای تجزیه شده حقیقی، انرژی قابل متابولیسم، ضریب تفکیک، pH و غلظت نیتروژن آمونیاکی در هر دو سوبسترا تحت تأثیر تیمارهای آزمایشی قرار نگرفت. نتیجهگیری نهایی: نتایج این تحقیق نشان داد که عملآوری کاه گندم و سرشاخه خرما توسط باکتریهای جداسازی شده سبب بهبود فراسنجههای تخمیر شکمبه در شرایط برونتنی گردید.

Introduction: There is a shortage of animal feed and water resources in several developing countries. Thus, proper use of agro industrial by products is a useful means to overcome this problem. Agricultural by products can use as animal feeds, but because of high lignin and low protein content, their digestibility and palatability is low. Several processing method has been used to improving nutritive value of such materials (Rahal et al. 1997 and Fazaeli et al. 2004). Recently, biological delignification of lignocellulosic material has been considered as an alternative approach (Fazaeli et al. 2004). Termites gut contains different kinds of lignocellulose degrading microbes, which the possibility of their use in improving the nutritive value of agricultural by-products is a major step in compensation of animal feed shortage.

Material and methods: The present study was conducted to determine the optimum temperature and pH for growth of symbiont bacteria degrading lignocellulosic material isolated from termite gut, and to investigate their effects on the digestibility and in vitro fermentation parameters of wheat straw (WS) and date leaf (DL). For this purpose, three bacteria including Bacillus licheniformis, Ochrobactrum intermedium and Microbacterium paludicola were used,which isolated by culture of termite guts contents in mediums containing different types of lignin and lignocellulosic materials. Sterile basal media was used as a medium for isolation process and contained the following ingredients per litre: 7.0 g K2HPO4, 3.0 g KH2PO4, 1.0 g (NH4)2SO4, 0.1 g MgSO4·7H2O (Borji 2003). Growth curves of isolated bacteria for optimum temperature and pH were obtained using nutrient broth medium. Three temperatures of 30, 37 and 40 oC and four pH of 5, 6, 7 and 8 were evaluated. After that, WS and DL were processed with these isolates in media 9 (M9) medium with following composition (per litre): 6.2 g Na2HPO4, 3.0 g KH2PO4, 0.5 g NaCl, 1.0 g NH4Cl (Kato et al. 1998). After processing, residues were collected, dried and used for in vitro gas production experiment according to Makkar (2010). For this purpose, rumen liquor was collected from two ruminally fistulated cows. Animals were maintained on a 60% hay and 40% concentrate diet according to their requirements for two weeks. Rumen contents were collected from the ventral sac of rumen before morning feeding. A completely randomized design was used to determine the optimum temperature and pH for bacterial growth as well as effects of different bacteria on in vitro gas production and fermentation parameters of treated substrates. Data were analyzed by General Linear Model (GLM) procedures of SAS (SAS Institute, 2001),

Results and discussion:All of isolates were capable of grow at all of examined temperatures (i.e., 30, 37 and 40 oC). The optimum growth of B. licheniformis occurred at 40 oC, but the optimum growth of O. intermedium and M. paludicola was observed at 37 oC. All of isolates grew at different pH ranges (i.e., 5, 6, 7 and 8) and their optimum growth was observed at neutral pH of 7.0. At this neutral pH, highest growth for B. licheniformis, O. intermedium and M. paludicola was obtained after 30, 34 and 36 h of incubation, respectively. After processing, in both WS and DL, highest and lowest gas production and organic matter (OM) digestibility were observed by B. licheniformis and control treatment, respectively (P<0.05). In vitro dry matter (DM) digestibility, truly digested substrate, metabolizable energy, partitioning factor, pH and ammonia nitrogen concentration were not affected by the experimental treatments. Factors such as temperature, pH, atmosphere gas composition, bacterial type, composition of media culture, number of bacteria used and end products bacteria and their interaction has an important effect on growth rate of isolates (Mohan and Manuselis 1995). Similar to our results, by isolation of Bacillus sphaericus, Ochrobactrum anthropi and Enterobacter cloacae from termite gut, Borji (2003) reported that their optimum temperature and pH were observed at 25-37 oC and 7-8, respectively. Other researchers also reported similar results (Giroux et al. 1988 and Yang et al. 1995). Totally, obtaining optimizing pH and temperature of isolated bacteria for processing agricultural by-products can maximize their nutritive value. Improving OM digestibility and fermentation parameters in both substrates was maybe due to effect of isolates on changing lignin structure and loosening its bounds with carbohydrate compounds (VanSoet 2004). Consistent to results of present study, borji (2003) indicated that inoculation of wheat straw and barley straw with different bacteria isolated from termite gut increased their DM digestibility compared to control treatment. Increasing DM and OM digestibility and volume of gas production in WS than DL was probably because of lower lignin and higher carbohydrate content in the former (Azizi-Shotorkhoft et al. 2016).

Conclusion:Results of present study showed that optimum growth for Ochrobactrum intermedium and Microbacterium paludicola isolated from termite gut was observed at 37 oC while it was 40 oC for Bacillus licheniformis. Optimum pH for growth of all of isolates was 7. Processing wheat straw and date leaf with bacteria, especially B. licheniformis improved rumen fermentation parameters in vitro. Research for upgrading the nutritive value of by products with ligninolytic bacteria isolated from termite gut is in infancy. Thus, further works in this field of study is needed.

بررسی امکان جایگزینی اسانس میخک به جای آنتیبیوتیک محرک رشد آویلامایسین در جیره غذایی جوجههای گوشتی
زمینه مطالعاتی: گیاهان دارویی به عنوان یکی از جایگزینهای مناسب برای آنتیبیوتیکهای محرک رشد مطرح هستند. هدف: این آزمایش به منظور بررسی امکان جایگزینی اسانس میخک به جای آویلامایسین و اثر سطوح مختلف آن بر عملکرد رشد، قابلیت هضم ایلئومی مواد مغذی، ریختسنجی مخاط ژژنوم و برخی فراسنجههای خونی انجام شد. روشکار:از تعداد 320 قطعه جوجه گوشتی راس 308 (مخلوط دوجنس) در قالب طرح کاملاً تصادفی استفاده شد. چهار تیمار آزمایشی شامل 10 قسمت در میلیون آویلامایسین (شاهد) و سه سطح 150، 300 و 450 قسمت در میلیون اسانس میخک، در چهار تکرار به 20 قطعه جوجه در هر تکرار به مدت 42 روز تغذیه شدند. نتایج: مصرف خوراک و افزایش وزن روزانه جوجهها در اثر مصرف اسانس میخک کاهش و ضریب تبدیل خوراک آنها در اثر سطوح 150 و 300 قسمت در میلیون افزایش یافت (05/0P<). استفاده از اسانس میخک باعث کاهش قابلیت هضم ایلئومی ماده خشک و ماده آلی جیره مصرفی شد (05/0P<). pH محتویات سنگدان و سکوم و نیز فراسنجههای ریختسنجی مخاط ژژنوم تحت تأثیر معنیدار تیمارها قرار نگرفتند. با افزودن سطوح مختلف اسانس میخک به جیره، غلظت کلسترول کل سرم خون جوجهها کاهش یافت (05/0P<) ولی افزایش غلظت HDL کلسترول و کاهش غلظت LDL کلسترول در اثر سطوح 150 و 450 قسمت در میلیون معنیدار شد (05/0P<). نتیجهگیری کلی: بهطور کلی اسانس میخک نمیتواند به جای آنتیبیوتیک محرک رشد آویلامایسین در جیره جوجههای گوشتی جایگزین شود و فقط میتواند برخی اثرات مفید را در کاهش کلسترول سرم خون نشان دهد.

بررسی غلظت مس، آهن و روی سرم خون اسب و قاطر در ارومیه
زمینه مقدماتی: تعیین مقادیر ریزمغذیها در خون روند رشد، تولید و تولیدمثل را در دامها نشان میدهد. در صورت مشاهده کمبود با اصلاح آنها میتوان از بروز بیماری جلوگیری کرد. هدف: تعیین مقادیر مس، آهن و روی سرم تکسمیهای ارومیه بر مبنای گونه، جنس، سن و تعیین ارتباط بین آنها. روش کار: مقدار 10 میلیلیتر خون از 100 راس نریان (43 راس) و مادیان (57 راس) اسب (78 راس) و قاطر (22 راس) از سنین 1 تا 21 سال از ورید وداج تهیه شد. مقادیر سرمی ریزمغذیها به روش جذب اتمی ارزیابی شدند. نتایج: میانگین غلظت سرمی مس، آهن و روی در تکسمیها بهترتیب 5/14، 2/40 و 48/7 میکروگرم در دسیلیتر بود. غلظت ریزمغذیها در قاطرها بیشتر از اسبها بود اما معنیدار نبودند. میانگین غلظت ریزمغذیها در نریان بیشتر از مادیان بود اما فقط آهن معنیدار بود (مادیانها کمتر از نریان)، مس و روی متفاوت نبودند. بالاترین و پایینترین غلظت سرمی مس بهترتیب در سنین 16 و 11 سالگی، آهن در 18 و 14 سالگی و روی در 16 و 14 سالگی بود. غلظت سرمی مس و آهن در سنین مختلف نزدیک به معنیدار (057/0P<) بود ولی روی متفاوت نبود. بالاترین و پایینترین غلظت سرمی ریزمغذیها بهترتیب در گروه سنی 21-13 و 6-1 سال بود که تفاوت معنیدار نبود. ارتباط مس/آهن (41/0=r)، مس/روی (92/0=r) و آهن/روی (47/0=r) مثبت و معنیدار بود که بالاترین ضریب همبستگی در مس/روی بود. نتیجهگیری نهایی: غلظت ریزمغذیها در تکسمیها در دامنه طبیعی بود. گونه، جنس و سن در میزان آنها موثر نیست مگر آهن که در مادیان کمتر از نریان بود و سنین 11 تا 14 سال در کمبود ریزمغذیها احتمالاً تعیینکننده باشد. لذا تکسمیهای ارومیه از نظر ریزمغذیها کمبودی نداشته اما آهن در مادیان با سن 14 سالگی بایستی مورد توجه باشد.

اثر مرحله بلوغ و زمان برداشت بر ارزش تغذیهای علوفه خشک یونجه برداشت شده از مناطق گرمسیر
زمینه مطالعاتی: عوامل موثر بر ارزش تغذیهای علوفه یونجه برای تغذیه نشخوارکنندگان. هدف: اثر مرحله بلوغ و زمان برداشت بر ارزش تغذیهای علوفه خشک یونجه برداشت شده از مناطق گرمسیری بررسی شد. روش کار: علوفه یونجه در دو مرحله بلوغ (قبل از گلدهی و اوایل گلدهی) و در دو زمان (صبح و عصر) برداشت شد. ترکیب شیمیایی، بخشهای مختلف پروتئین خام، پروتئین غیر قابل تجزیه در شکمبه و تجزیهپذیری شکمبهای ماده خشک، ماده آلی، پروتئین خام و الیاف شوینده خنثی تعیین شد. نتایج: علوفه یونجه برداشت شده در اوایل گلدهی در مقایسه با قبل از گلدهی دارای الیاف، لیگنین، پروتئین غیر قابل تجزیه در شکمبه، عصاره اتری و بخش B3 و C پروتئین بیشتر و کربوهیدراتهای غیر الیافی و تجزیهپذیری شکمبهای ماده خشک و ماده آلی کمتری بود (05/0>P). محتوای بخش A پروتئین، پروتئین خام، کلسیم و منیزیم علوفه خشک یونجه تحت تاثیر مرحله بلوغ قرار نگرفت. برداشت یونجه در صبح در مقایسه با عصر محتوای پروتئین خام، الیاف، پروتئین غیر قابل حل در شوینده خنثی و اسیدی و پروتئین غیرقابل تجزیه در شکمبه را افزایش و محتوای کربوهیدراتهای غیرالیافی و تجزیهپذیری شکمبهای ماده خشک و ماده آلی را کاهش داد (05/0>P). محتوای بخش B3 پروتئین، ماده آلی، عصاره اتری، لیگنین و کلسیم تحت تاثیر زمان برداشت علوفه یونجه قرار نگرفت. نتیجهگیری نهایی: یونجه برداشت شده در قبل از گلدهی و نوبت عصر ارزش تغذیهای بالاتری داشت.

بررسی ناحیه HVR-III ژنوم میتوکندری گوسفندان ایرانی با روش توالییابی
زمینه مطالعاتی: کشور ایران جزء یکی از محدود مناطقی است که کشاورزی و اهلی شدن حیوانات توسط انسانهای اولیه در آن اتفاق افتاده است و در نتیجه باعث شده است که ایران دارای ذخایر ژنتیکی ارزشمندی در حیوانات اهلی باشد. از طرفی ایران با داشتن بیش از 27 نژاد گوسفند اهلی به همراه 5 زیرگونه گوسفند وحشی، دارای ذخیره ژنتیکی با تنوع کم نظیری در دنیا می باشد. هدف: هدف از انجام این تحقیق توالییابی ناحیه HVR-III از ژنوم میتوکندری گوسفندان اهلی نژاد مغانی، لریبختیاری، سنگسری، بلوچی، کرمانی، شال، قرهگل و کردی به منظور بررسی ارتباط فیلوژنیک بین نژادهای مذکور و همچنین مقایسه با هاپلوتایپهای شاخص شناسایی شده گوسفند اهلی میباشد. روش کار: برای این منظور تعداد 80 نمونه خون جمعآوری و پس از استخراج DNA، ناحیه موردنظر توسط پرایمرهای اختصاصی به روش PCR تکثیر و سپس توالییابی شدند. نتایج: با تجزیه و تحلیل دادههای حاصل از تعیین توالی و مقایسه توالی HVR-III گوسفندان ایرانی با یکدیگر و با توالی ناحیه مشابه از ژنوم گوسفندان سایر نژادها، مشخص شد که علیرغم تنوع فنوتیپی قابل توجه در بین نژادهای مورد مطالعه، این نژادها همگی متعلق به گروه هاپلوتایپی A میباشد. این امر ممکن است به دلیل نزدیکی جغرافیایی مکان زیستی آن ها و یا به دلیل روش خاص زندگی عشایر باشد که انتظار می رود نژادهای متنوع امروزی ایران از توده های محدودی با تنوع نسبتا کم مشتق شده باشند. نتیجه گیری نهایی: نتیجه گیری کامل تر نیازمند مطالعه بخشهای بزرگتری از DNA میتوگندری و همچنین بر روی تعداد بیشتر نمونه از نژادهای اهلی به همراه مطالعه تنوع نژادهای وحشی گوسفند ایران است.

زمینه مطالعاتی: در سالهای اخیر استفاده از روغنها در جیره دام به دلیل داشتن برخی اسیدهای چرب مفید برای سلامتی انسان همچون اسیدلینولئیک و اسیدلینولنیک مورد توجه قرار گرفته است. هدف: بررسی تاثیر افزودن روغنهای کلزا، سویا و ماهی به جیره خوراکی بر کیفیت و ترکیب اسیدهای چرب گوشت برههای پرواری بود. روش کار: 35 راس بره نر با میانگین وزن 2 ±8/27 کیلوگرم در یک دورهی 84 روزه مورد استفاده قرار گرفت. تیمارهای آزمایشی شامل 1) شاهد (بدون افزودن روغن)، 2) 3 درصد روغن ماهی، 3) 3 درصد روغن کلزا، 4) 3 درصد روغن سویا، 5) 5/1 درصد روغن ماهی + 5/1 درصد روغن کلزا، 6) 5/1 درصد روغن ماهی + 5/1 درصد روغن سویا و 7) 5/1 درصد روغن کلزا + 5/1 درصد روغن سویا بودند. نتایج: منابع مختلف روغن در سطح 3 درصد اثر معنیداری بر رنگ،pH نهایی، شاخص افت حاصل از پخت و نیروی برش گوشت نداشتند. اسیدهای چرب اولئیک، پالمیتیک و استئاریک به ترتیب بشترین مقدار را در بین تمام اسیدهای چرب گوشت داشتند. جیرههای حاوی روغن سبب افزایش میزان اسیدهای چرب غیراشباع و کاهش اسیدهای چرب اشباع در برههای تغذیه شده با روغن در مقایسه با تیمار شاهد شدند. برههای تغذیه شده با روغن ماهی بالاترین میزان اسیدهای چرب ایکوزاپنتانوئیک، دوکوزاپنتانوئیک و دوکوزاهگزانوئیک را داشتند. افزودن روغن به طور موثر نسبت اسیدهای چرب امگا-6 به امگا-3 را در مقایسه با شاهد کاهش داد. نتیجهگیری نهایی: افزودن منابع مختلف روغن به جیره دام، میتواند بدون اثرات منفی بر کیفیت گوشت، ترکیب اسیدهای چرب گوشت برهها را تحت تاثیر قرار دهد و به عنوان یک راهکار جهت افزایش اسیدهای چرب غیراشباع گوشت، بخصوص اسیدهای چرب امگا-3 و بهبود نسبت امگا-6 به امگا-3 استفاده شود.

تاثیر تزریق گونادوتروپین کوریونیک انسانی پس از قوچاندازی بر غلظت پروژسترون و عملکرد تولیدمثلی میشهای آمیخته افشاری × برولا مرینو
زمینه مطالعاتی: تلفات اولیه رویانی یکی از مشکلات اصلی تولیدمثلی دامهای مزرعهای است. تولید ناکافی پروژسترون از جسم زرد در اوایل آبستنی یکی از علل عمده این تلفات است. هدف: هدف از انجام این آزمایش تعیین اثرات تزریق عضلانی 400 واحد بینالمللی hCG، در روز 11 پس از قوچاندازی بر روی عملکرد تولیدمثلی میشهای آمیخته افشاری × مرینو بود. روش کار: در مطالعه حاضر، تعداد 24 راس میش آمیخته افشاری × مرینو به صورت تصادفی در دو گروه 12 راسی قرار گرفتند. جهت همزمانی فحلی، تمامی میشها سیدرهای پروژسترونی را به مدت 12 روز دریافت کردند. در روز 11 پس از قوچاندازی، میشهای گروه hCG، 400 واحد بینالمللی hCG به صورت عضلانی دریافت کردند و میشهای گروه کنترل هیچ درمانی را دریافت نکردند. جهت اندازهگیری غلظت هورمون پروژسترون در روزهای 7، 12، 17 و 22 پس از قوچاندازی از میشها خونگیری به عمل آمد. نتایج: نتایج این مطالعه نشان داد که غلظت هورمون پروژسترون در روزهای 12 و 17 پس از قوچاندازی در میشهای گروه hCG به طور معنیداری بالاتر از گروه کنترل بود (05/0 >P). میشهای گروه hCG سه و چهارقلوزایی بیشتری نسبت به گروه کنترل داشتند که در نتیجه تعداد بره متولد شده و تعداد بره به ازای هر میش در گروه hCG بالاتر بود. میزان تلفات رویانی تا زمان از شیرگیری در گروه hCG (6/3%) از گروه کنترل (8/11%) کمتر بود (05/0>P). نتیجهگیری نهایی: به طور کلی نتایج بدست آمده از این تحقیق نشان داد که تجویز hCG در اوایل آبستنی با بهبود عملکرد لوتئال و احتمالا با کاهش تلفات رویانی، عملکرد تولیدمثلی را در میشهای آمیخته افشاری × مرینو بهبود میبخشد.

اثر افزودنی باکتریایی و پریبیوتیکی بر پروفایل تخمیر و تجزیهپذیری شکمبهای مواد مغذی سیلاژ ذرت
زمینه مطالعاتی: استفاده از برخی مواد افزودنی به سیلاژ ذرت میتواند بر پروفایل تخمیر و تجزیهپذیری شکمبهای مواد مغذی سیلاژ ذرت تاثیر بهسزایی داشته باشد. هدف:این طرح به منظور بررسی اثرات افزودنی میکروبی تجاری با نام Lalsil Fresh (حاوی لاکتوباسیلوس بوکنری) و افزودنی پریبیوتیکی (پودر آب پنیر) بر روی پروفایل تخمیر و تجزیهپذیری شکمبهای مواد مغذی سیلاژ ذرت صورت گرفت. روش کار: تیمارهای آزمایشی بترتیب شامل 1) سیلاژ ذرت شاهد (بدون افزودنی)، 2) سیلاژ ذرت تیمار شده با پودر آب پنیر (یک درصد یا 10 کیلو بر تن)، 3) سیلاژ ذرت تیمار شده با افزودنی باکتریایی Lalsil Fresh به میزان 106×8/1 واحد تشکیل دهنده کلنی به ازای هر گرم علوفه تازه و 4) سیلاژ ذرت تیمار شده با پودر آب پنیر (یک درصد) به همراه افزودنی باکتریایی لالسیل (106×8/1 واحد تشکیل دهنده کلنی به ازای هر گرم علوفه تازه) بودند. نتایج: در پایان دوره آزمایشی 90 روزه تیمار پودر آب پنیر و افزودنی باکتریایی سبب کاهش تولید پساب و افزایش در ماده خشک سیلاژ نسبت به تیمار شاهد شدند (05/0< P). افزودن پودر آب پنیر به علوفه ذرت سیلوئی سبب افزایش غلظت پروتئین خام و کاهش غلظت الیاف در سیلاژ گردید (05/0< P). افزودنی لالسیل باعث کاهش pH سیلاژ ذرت شد (05/0< P). مخلوط افزودنی میکروبی و پریبیوتیکی کمترین میزان اسیدهای چرب فرار و ازت آمونیاکی را به خود اختصاص دادند (05/0< P). تیمار پودر آب پنیر سبب افزایش معنیدار تجزیهپذیری بخش محلول، بخش کند تجزیه و ثابت نرخ تجزیه گردید (05/0< P). افزودنی باکتریایی نیز سبب افزایش قابلیت ناپدید شدن ماده خشک سیلاژ ذرت شد. نتیجهگیری نهایی: نتایج آزمایش نشان داد که پودر آب پنیر میتواند سبب بهبود ارزش تغذیهای، افزایش قابلیت هضم و نرخ نجزیه پذیری سیلاژ ذرت گردد. همچنین افزودنی باکتریایی با کاهش سریع pH موجب بهبود کیفیت سیلاژ حاصله میگردد.

اثراسانس گیاه نعناع فلفلی (Mentha piperita) بر لاکتوباسیلهای جدا شده از شکمبه گاو
زمینه مطالعاتی: اسانس برخی گیاهان فعالیت ضد میکروارگانیزمی دارد. هدف: این تحقیق به منظور بررسی اثر اسانس گیاه نعناع فلفلی بر لاکتوباسیلهای جداسازی شده از شکمبه گاو به منظور مطالعه امکان کنترل اسیدوز از طریق کاهش این میکروارگانیزمها انجام شد. روش کار: نمونه مایع شکمبه از شکمبه گاوهایی که با جیره حاوی 65%کنستانتره و 35 % علوفه تغذیه شده بودند جمعآوری و صاف گردید. بهمنظور جداسازی لاکتوباسیلها در سطح جنس از محیط اختصاصی MRS استفاده گردید و انتخاب کلنیها بر اساس اندازه و مورفولوژی و تستهای کاتالاز و اکسیداز و رنگآمیزی گرم انجام گرفت. سپس تستهای بیوشیمیایی تخمیر قندها و هیدرولیز آرژنین و تولید CO2از گلوکز انجام شد. به منظور بررسی تأثیر اسانس نعناع فلفلی بر لاکتوباسیلهای جداسازی شده و لاکتوباسیلوس پلنتارم ابتدا اسانس گیاه بوسیله دستگاه کلونجر استخراج شد و سپس اثر آن به کمک روش ایجاد حلقه در آگار و اندازهگیری قطر هاله عدم رشد انجام گرفت. آزمایش در قالب طرح کاملاً تصادفی با سه تکرار انجام شد. تیمارهای آزمایش شامل 16 گونه لاکتوباسیلوس بود که در معرض غلظت ثابتی از اسانس خالص نعناع فلفلی وآب مقطر (شاهد) قرار گرفتند. نتایج: تعداد 36 کلنی مربوط به جنس لاکتوباسیلها جداسازی و شناسایی گردید که در15گروه در سه طبقه تقسیم بندی شدند. لاکتوباسیلهای هتروفرمنتیتیو با 58% فراوانی بیشتری نسبت به لاکتو باسیلهای هموفرمنتیتیو با 42% داشتند. اسانس نعناع بر تمام باکتریهای آزمایش شده اثر مهاری داشت. نتیجه گیری نهایی: اسانس گیاه نعناع فلفلی اثر معنیداری (05/0p≤) بر کاهش رشد لاکتوباسیلها در محیط آزمایشگاه داشت.
